Cloning, sequencing, and expression of the gene encoding an endo-beta-1,4-xylanase from Myceliophthora heterothallica and Myceliophthora thermophila, and characterization of the recombinant enzyme
As global markets for industrial enzymes grow substantially in the discovery of new enzymes and applications, xylanases have been widely used in an extensive range of industrial processes. In the enzyme industry, applications of thermostable enzymes from thermophilic fungi have been a potential field for research, due to advantages from their elevated hydrolysis temperatures, at which carbohydrate is broken into sugars. However, little has been known about the activities of enzymes that enable their elevated hydrolytic power, as well as the genomes of these thermophilic fungi. By cloning, sequencing, and testing the gene expression of two thermophilic Myceliophthora strains, I hoped to confirm the sequences of the predicted genes in our experimental strains, which were assumed to encode an endo-β-1,4-xylanase, as well as the characterization of the predicted recombinant enzymes.
